Dear friend, here at USPlabs we love BCAAs. The problem is that while taking them, we are simultaneously consuming artificial flavors and dyes in large quantities, which gave us headaches, nausea, lethargy and, frankly, the taste of shit in our mouths. Also, and perhaps most importantly, we just couldn't find BCAAs with the amino acid ratio we wanted.

The Little-Known mTOR Anabolic Pathway If you've never heard of mTOR (and even if you have), listen carefully and listen well, as it's one of the highest rated anabolic protein synthesis factors known to man. And most likely, you were not even close to the maximum of your huge potential.
mTOR (mammalian target of rapamycin) is a proline-directed serine-threonine protein kinase responsible for a wide range of physiological and metabolic functions in several cell types, including both adipocytes (fat cells) and myocytes (muscle cells).
Among its many functions, mTOR is responsible for hypertrophic growth (eg, muscle cell enlargement), protein synthesis, and tissue morphogenesis.As a result, components that aim to regulate mTOR production have been sought after for quite some time. A wide range of evidence suggests that amino acids are able to directly regulate mTOR signaling pathways in skeletal muscle, including the possibility that they upregulate some of the mTOR-dependent insulin/growth factor-enhancing cellular signaling elements. However, the same data show that amino acids do not uniformly activate mTOR signaling pathways, and there is evidence that the mechanism of mTOR activation can be either the entire amino acid matrix or one specific amino acid. In particular, the amino acid leucine is indicated as the main one responsible for the activation of branched chain amino acids causing the activation of mTOR signaling pathways and therefore the mTOR effect, on the contrary, triggers protein synthesis. This hypothesis arose because studies have shown that most of the effects of amino acids on mTOR signaling are reversed with a decrease in leucine concentration or increased with the addition of leucine and, to a lesser extent than other branched chain amino acids.
In other words, leucine is the main one, but it should also be mentioned that the presence of two other amino acids – isoleucine and valine – is necessary in order to maximize the result.

8-1-1 BCAA Ratio With that said, we've compiled USPlabs Modern BCAAs with an 8-1-1 ratio: Leucine-Isoleucine-Valine.

Results and Meanings As you know, many people double, triple, and even quadruple their BCAA dosage in 2:1:1 and 4:1:1 ratios to achieve the best results.Given the fact that leucine is the most responsible amino acid for growth effects, especially in skeletal muscle, having an 8:1:1 formula eliminates the need for such increased doses. This provides an additional benefit along with other benefits.

Another problem is amino acid transport and metabolism. All three branched chain amino acids, leucine, isoleucine and valine, compete for the same transport systems. And as a result, the presence of high concentrations of the other two chained amino acids can lower intracellular levels of leucine. Obviously this is not what you want. This is further exacerbated by the fact that once leucine is ingested, it first assumes other structural roles unrelated to protein metabolism prior to activation of the mTOR signaling pathway. And as a result, BCAAs at a ratio of 2:1:1 or 4:1:1 actually counteract the anabolic effects of the product, as these substrates are pitted against each other in competition for metabolism.

Sustamine
This product consists of a glutamine dipeptide (L-alanyl-L-glutamine) which is more stable and much more water soluble than regular L-glutamine. These physico-chemical properties alone can provide greater efficacy than conventional L-glutamine. However, the presence of a unique intestinal transporter in the dipeptide may also allow an increase in the amount of glutamine in the bloodstream and in skeletal muscle compared to normal free L-glutamine. With the introduction of alanyl-glutamine, a higher level of glutamine concentration in the blood plasma and skeletal muscles of animal models was maintained, compared with the introduction of free L-glutamine.This is very important as up to 65% of regular L-glutamine can be destroyed before it makes its way to the muscles.
